Kinetics of Cytotoxicity and Proliferation

To determine kinetics of compound potency and efficacy based on live-cell imaging.​

Direct Monitoring of Cell Number
Classic viability assays rely on the proportionality between cell number and a cell's functional activity or a cellular marker, as MTT assay. If a treatment affects the same functional activity or marker levels used to estimate cell number, the outcome of the assay will be representative of the changes in that biomarker, not of cell number. For instance, this limitation is of relevance for compounds which affect metabolic activity, as many anticancer agents. In contrast, our assay let to directly count exact number of cells and its outcome will be truly representative of cell viability.
Evaluation of Phenotypic Effect
Classic viability assays do not discriminate between arrest of cell proliferation and induction of cell death. However,  best candidates are the ones that kill tumor cells, not simply reduce their growth, and more assays are needed to assess the cytotoxic effect of tested compounds. In contrast, our assay let to clearly discriminate between reduction of proliferation and a cytotoxicity, since direct separate count of live and dead cells is performed. Save efforts, time and money by picking the right candidates one step before.
Identify timing of drug exposure
Classic viability assays provide a single end-point measurement of treatment potency. However, two compounds can exert similar potency after 48 hours of treatment but one of them could require less time of exposure to induce its effects. In combination with pharmacokinetics studies, this information would be of extreme relevance to decide how to continue the project. Our assay let to assess compound potency up to every 15 minutes for 48 hours of treatment and provide a detailed kinetics of potency and efficacy.
